Some of those blocks are way over a ton, so whatever the mold was made of, it would have had to be very tough (to withstand the pressure bearing down in the mold) and also stiff (so that it doesn't bulge too far out). Check out the large 280 ton block at Sacsayhuaman. At least some of those larger ground-level blocks might very well be shaped bolders that were lying around, but notice the different shade of the blocks sitting on top of the larger ground blocks:

When I visited there it was not possible to slip a business card in the joints. I can't fathom how such things could have been carved from solid rock and fitted into that kind of non-linear complex geometry.
I believe the closest thing we have to a true smoking gun is the Wall of the Six Monoliths at Ollantaytambo. The entire enigma of that wall can be explained by the "pour and cast" paradigm, including why the bosses are on the side, why one side of each large slab is flat while the other side is curved, and why there are thin "spacer" stones (with a "boss" at the bottom of each) between those larger blocks.
On big problem though: we don't know what the casts could have been made of, nor do we know what was actually poured into them!
